This mouse IgG1 antibody was generated against human activated tonsillar B cells and is specific for human CD40.
B cells require a series of coordinated signals in order to undergo maturation including MHC antigen presentation, cytokine stimulation (IL-2/4/5) and costimulation. CD40 is a costimulatory protein found on antigen presenting cells (such as B Cells) and is required for their activation. The binding of CD154 (CD40L) on T helper cells to the CD40 receptor stimulates antigen presenting cells to undergo maturity and develop downstream effector functions.

Immunopreclpltatlons wlth detergent extracts of 125I-labeled tonsillar B cells. Lysates were treated with complexes of goat anti-mouse with anti-CD40-626 mAb and goat anti-mouse Ig. The precipitates were run on a 10% SDS-PAGE under reducing conditions. Control Ig mAb HDP-1 (lane a) and anti-CD40-626 (lane b).
