Dde Biotin-DBCO is an azide-reactive biotin probe that allows for efficient release of the captured biomolecules from streptavidin. This probe contains a biotin moiety linked to an DBCO group through a hydrazine cleavable Dde linker. Captured biomolecules can be efficiently released under mild aqueous conditions with hydrazine.
Extraordinary strength of the streptavidin-biotin interaction allows for efficient capturing of even highly dilute targets; however, it makes recovery of proteins from affinity resins challenging. This probe eliminates a major limitation of the streptavidin-biotin affinity purification.
