AA16 recognizes the human Class VI Beta Tubulin isotype. The antibody does not recognize any other Beta Tubulin isotype, and it does not react with the rodent Class VI.
Class VI Beta Tubulin (Tubb1) is present exclusively in megakaryocytes and their derivatives, platelets. Transgenic mice lacking Class VI have non-discoid platelets with defective marginal bands, and suffer from thrombocytopenia and excessive bleeding.

Pattern of reactivity of Anti-?-Tubulin antibodies against MBP-C-terminal ?-Tubulin fusion proteins. The following MBP-C-terminal ?-Tubulin fusion proteins were spotted: Class I ?-Tubulin, Class II ?-Tubulin, Class III ?-Tubulin, Class IVa ?-Tubulin, Class IVb ?-Tubulin, Class V ?-Tubulin, Class VI ?-Tubulin. Antibody for Class III ?-Tubulin, Class VI ?-Tubulin (AA16), or none was applied to spots. Figure courtesy of: A. Frankfurter and A. Spano (unpublished data). |
