This mouse monoclonal antibody was raised against recombinant GST-nonstructural 5B protein (NS5B), aa 1-346, Hepatitis C virus (HCV) genotype 2a strain fusion protein and recognizes NS5B of HCV.
Highlights:
Hepatitis C is an infectious disease caused by the hepatitis C virus (HCV) that primarily affects the liver. HCV encodes a polyprotein that is cleaved into at least 10 viral proteins namely E1, E2, core, p7, NS2, NS3, NS4A, NS4B, NS5A, and NS5B. Non-structural 5B protein (NS5B) is HCV's RNA dependant RNA polymerase and is critical for viral replication.

Immunofluorescence
Huh7.5 cells were mock infected or infected with HCV were fixed using 4% formaldehyde solution at 2 days post infection. Immunofluorescence assay was performed using purified 1A4 at the 20ug/ml. DAPI was used to stain the nucleus.
Western Blot
Genes encoding full length NS5B proteins (with a N-terminal flag-tag) from 3 HCV genotypes, namely 1b, 2a, and 3a, were over-expressed in 293FT cells, followed by lysis of cells in RIPA buffer, Mock refers to cells transfected with empty vector only. SDS-PAGE separation and transfer by Western blotting. Membranes were probed with purified mAbs at 2ug/ml.
