LivePAR lentivirus preparation for expression of a genetically encoded poly-ADP-ribose (PAR) monitor in mammalian cells.
To overcome limitations of immunocytochemistry for poly-ADP-ribose (PAR) analysis in cells, the Sobol lab has developed a lentiviral vector for expression of a fluorescently tagged PAR-binding domain (PBD) fusion protein: LivePAR, a Live-cell imaging probe for poly(ADP-Ribosyl)ation (PAR). Studies from the Sobol lab document that the LivePAR probe is highly sensitive for PAR detection in cells, revealing the temporal dynamics of PAR formation and degradation by confocal fluorescent microscopy.
