MCF-7 cells were treated with an inhibitor of mitochondrial DNA (mtDNA) replication for 69 days, after which surviving cells were cloned, the resulting clones were analyzed for mtDNA content by PCR, and the cells' identity was confirmed by STR analysis. Clone #5d lacks mitochondrial DNA (rho-0; ρ-0) and can be used as a recipient of mtDNA. Its STR profile matches that of Cellosaurus MCF-7 (CVCL_0031).
