Mouse fibroblast line lacking the Risk Iron Sulfur Protein from complex III of the respiratory chain (RIESKE protein).
Rieske proteins are iron-sulfur protein (ISP) components of cytochrome bc1 complexes and cytochrome b6f complexes and responsible for electron transfer in some biological systems. To generate the cell line, primary lung fibroblast culture was stablished from a knockin mouse homozygous for the floxed UQCRFS1 (exon 2) gene. UQCRFS1 encodes for the Rieske iron sulfur protein, subunit of complex III. Cells were immortalized by retroviral transduction with E6/E7 type 16 papiloma virus carrying G418 resistance. Clones were obtained from immortalization. One of the clones #8 was used to generate the RIESKE KO cells by transfecting with a plasmid expressing cre recombinase. Clones were obtained after hygromycine selection. This is KO clone is #8.5. Generation and characterization of cell line is described in PMID: 22106410. Mouse used to generate cell line had mixed genetic background (129 svj and C57BL6J).
